SYLLABUS

1) Historical aspect of plant cell, tissue and organ culture.
2) Plant tissue culture laboratory, aseptic techniques and nutritional components of tissue culture medium
3) Initiation of callus.
4) Maintenance of callous, organ genesis
5) Plant propogation through tissue culture, micropropogation of flowering plants, commercial application with reference to floriculture industry.
6) Somatic embryogenesis
7) Synthetic seeds. Commercial potentials of synthetic seeds
8) Plant protoplasts: Isolation, culture and somatic hybridization
9) Somaclonal variation
10) Hardening process
11) Plant tissue culture industry in India.
12) Application of tissue culture in agriculture, forestry and horticulture


PRACTICALS

1) Introduction to plant tissue culture: Laboratory, instruments, glasswares and their handling
2) Various sterilization techniques.
3) Preparation of stock solutions.
4) Preparation of MS medium.
5) Seed sterilization and in-vitro germination.
6) callus induction from suitable explant material.
7) Auxiliary bud proliferation and regeneration of plantlets (any suitable material)
8) Encapsulation of auxiliary buds.
9) Maintenance of callus culture.
10) Shoot multiplication.
11) Hardening of plants.

Projects - Micropropogation of any one medicinal plant / ornamental plant / Floriculture plant / Fruit crop / Economically important plant.
